Abstract:Objective To explore the feasibility of using a microfluidic PDMS chip punch in the preparation of multi-tissue external controls for immunohistochemistry and the advantages of promoting it in grassroots hospital pathology departments. Methods We reviewed breast cancer cases from the pathology department of Jiangyin Traditional Chinese Medicine Hospital from 2021 to 2024. After pathological diagnosis, we selected paraffin blocks with residual tumor tissues showing HER2 protein expression levels of 0, 1+, 2+, and 3+. Tissue strips were sampled using a PDMS chip punch and embedded to create a HER2 multi-tissue external control composite block. Results The HER2 multi-tissue controls prepared using the PDMS chip punch were attached to the same slide as the test tissues. After immunohistochemical staining, the control tissues were neatly arranged, saved space, had accurate positive localization, clear background, and were convenient for viewing. Conclusion This study presents a simple and practical method for preparing multi-tissue external controls for immunohistochemistry. The PDMS chip punch used is low-cost and easy to operate, making it highly suitable for promotion in grassroots hospital pathology departments, providing an important basis for immunohistochemical quality assessment.
