Comparison of effects of modified intraoperative frozen section fixation method in HE staining

Abstract:Objective To investigate the differences in HE staining effects among different fixation methods for frozen sections. Methods Thirty fresh specimens remaining after intraoperative rapid pathological diagnosis at the Department of Pathology, Guoyao Dongfeng General Hospital, from October to November 2024 were collected. Frozen sections were fixed using three different methods: Group A (single fixation), Group B (delayed drying fixation), and Group C (multiple immediate fixation). The HE staining results of the three groups were compared. Results The frozen sections prepared using the multiple immediate fixation method scored higher in four indicators: tissue structure, nuclear staining intensity, cytoplasmic staining intensity, and nuclear-cytoplasmic contrast, with statistically significant differences (P<0.05). Conclusion The multiple immediate fixation method can significantly improve the quality of HE staining in frozen sections.
