Abstract:Objective: To explore the more suitable dehydration procedures for needle biopsy specimens using needle biopsy embedding boxes. Methods: A total of 60 needle biopsy specimens were selected from the People's Hospital of the Xinjiang Uygur Autonomous Region, including 30 liver biopsy specimens and 30 breast biopsy specimens. The differences in the processing effects of tissue specimens using needle biopsy-specific embedding boxes combined with modified dehydration procedures versus traditional dehydration procedures were compared in terms of the ease of sectioning, HE staining, special staining, and immunohistochemical staining. Results: Compared to the conventional dehydration procedure, the use of needle biopsy-specific embedding boxes combined with a modified dehydration procedure resulted in easier sectioning, better quality of HE staining, immunohistochemical staining, and special staining. Conclusion: Using needle biopsy-specific embedding boxes not only allows the fixative to better penetrate the needle biopsy tissue but also promotes liquid exchange between various reagents and the biopsy tissue. Combined with the modified dehydration procedure, the resulting tissue is appropriately soft and hard, allowing for continuous and complete sectioning, with well-preserved antigens, and the quality of immunohistochemical and special staining preparations is excellent.
