Impact of KRAS/TP53 co-mutation on the therapeutic efficacy and prognosis of colorectal cancer patients

Abstract:Colorectal cancer is one of the most common malignant tumors in humans. During the occurrence and development of colorectal cancer, KRAS and TP53 play key roles. KRAS gene mutations can drive abnormal proliferation signals, while TP53 gene mutations disrupt genomic stability and promote tumor development. These two genes are the most common mutations in colorectal cancer, and KRAS/TP53 co-mutations can lead to more aggressive colorectal tumors and increase drug resistance. Their mutation status is of great significance for prognosis evaluation and treatment selection in colorectal cancer, and they are also a focus of research on new targeted therapies. This article introduces the basic functions of KRAS and TP53 and their mutation characteristics in colorectal cancer, and summarizes the impact of KRAS/TP53 co-mutations on the efficacy and prognosis of colorectal cancer patients, as well as existing targeted therapy strategies for KRAS/TP53 co-mutations, hoping to provide new insights for the clinical treatment of colorectal cancer.
