Expression and prognostic features of ACSM 1 and LIF in prostate cancer

Abstract:Objective To investigate the expression and significance of ACSM1 and LIF in prostate carcinoma(PCa).Methods The String and GEPIA2 databases were used to screen out genes related to AR based on the combined TCGA+GTEx database,and box plots showing the expression differences between prostate cancer tumor tissues and the corresponding normal tissues were obtained.The relevant clinical information of PCa patients was extracted from TCGA+GTEx,and the cases were divided into high-risk and low-risk groups to evaluate the prognostic value of LIF and ACSM1.Pathological paraffin blocks from 75 patients were selected and analyzed for the expression of AR,LIF,and ACSM1 using immunohistochemical staining.Results Two genes,LIF and ACSM1,which interact with AR were screened out.The expression of ACSM1 and LIF in prostate cancer tissues was higher than those in normal tissues in the study(P<0.05).In AR-positive prostate cancer cases,the high expression of ACSM1 and LIF was positively correlated with tumor grade and prognosis(P<0.05).The positive expression of ACSM1 was positively correlated with Syn.The combined database showed that the prognosis of PCa patients in the low-expression group of ACSM1 and LIF was better than that in the high-expression group,with significant differences in OS and PFS.Conclusion Both ACSM1 and LIF are highly expressed in prostate cancer,and the patients with positive expression have a poor prognosis.ACSM1 may act in concert with LIF in PCa,providing new insights for clinical application.
