Expression of EFNA1 in cervical cancer and its effect on cancer cell activity

Abstract:Objective To investigate the expression of EFNA1 in cervical cancer and its effect on cancer cell viability.Methods The GEPIA database was used to analyze the expression of EFNA1 in cervical cancer tissues and adjacent tissues,as well as its correlation with the survival of patients.Cervical cancer tissues and adjacent tissues were collected from 47 patients with cervical cancer diagnosed and treated in our hospital from December 2021 to January 2025.The expression of EFNA1 in cervical cancer tissues and adjacent tissues was analyzed by immunohistochemistry and Western blot.Cervical cancer SiHa cells were divided into three experimental groups:siRNA NC group,siRNA EFNA1 group and KYA17971K group.EDU staining was used to analyze the proliferation activity of SiHa cells.Transwell assay was used to analyze the invasion ability of SiHa cells.The protein expressions of EFNA1,Wnt3α and β-catenin in SiHa cells were analyzed by Western blot.Results The analysis results of GEPIA database showed that EFNA1 was highly expressed in cervical cancer tissues,and the high expression of EFNA1 was associated with poor prognosis of cervical cancer patients.The results of this study showed that the expression of EFNA1 was increased in cervical cancer tissues compared with adjacent tissues(P<0.05).Compared with the siRNA NC group,the proliferation ability of SiHa cells in the siRNA EFNA1 and KYA17971K groups was decreased(P<0.05).The invasion ability of SiHa cells treated with siRNA EFNA1 and KYA17971K was significantly decreased(P<0.05).The expression of Wnt3α and β-catenin protein in SiHa cells treated with siRNA EFNA1 and KYA17971K decreased(P<0.05).Conclusion EFNA1 expression is up-regulated in cervical cancer tissues.Downregulation of EFNA1 expression can reduce the proliferation and invasion of cervical cancer SiHa cells,which is related to the regulation of Wnt/β-catenin pathway by EFNA1.
