The application of autopsy tissue in multi-tissue control system

Abstract:Objective To explore the application of autopsy tissues in the multi-tissue control quality assurance system. Methods After routine biopsy sampling, the remaining tissues including tonsil, thyroid, placenta, uterine myometrium, amnion were selected from the Department of Pathology of Xinjiang Uygur Autonomous Region People's Hospital. These tissues were combined with autopsy tissues (brain, liver, spleen, lung, kidney, stomach, pancreas) sent by Xinjiang Hengcheng Judicial Appraisal Institute. After 24-48 hours of additional fixation, all tissues were cut into 0.3 cm × 3 cm strips and tightly wrapped with amnion to form a multi-tissue amnion roll control paraffin block. This was applied in HE staining, special staining, and immunohistochemical quality control. Results In HE staining quality control, when reagents were fresh, all tissues showed clear cell nuclei and bright cells. When reagent conditions were poor, all tissues showed gray and dark staining of cell nuclei and cytoplasm, with reduced nuclear-cytoplasmic contrast. In special staining quality control, reticular fibers in liver tissue stained black in reticulin staining, collagen fibers stained blue in Masson staining, while others stained red, showing bright and clear staining. PAS staining showed purple-red staining of the basement membrane of glomerular capsule and tubules in renal tissue, and blue-black staining of elastic fibers in pulmonary tissue. In immunohistochemical quality control, the multi-tissue control could cover most antibody expressions of positive, weakly positive, and negative. Conclusion Applying autopsy tissues to the preparation of multi-tissue control paraffin blocks not only shortens the specimen collection cycle but also meets the quality control requirements for various staining methods, ensuring the accuracy of experimental results.
