Improvement of PASM staining method for kidney biopsy

Abstract:Objective To explore a better experimental protocol for PASM staining in kidney biopsy to improve the quality and efficiency of daily laboratory work.Methods A total of 30 cases of membranous nephropathy,amyloidosis nephropathy and membranous proliferative glomerulonephritis were collected from the renal biopsy and puncture specimens of Nanjing Jinyu Medical Laboratory Co.,Ltd.in 2024,and the classical method and the modified method were used to compare the differences in staining effect,diagnostic results,production efficiency and background.Results ①The staining effect of the two methods was satisfactory,the basement membrane was black,the nucleus was blue,the cytoplasm was red,and the tissue was brightly colored and contrasted.② The results of staining pathological diagnosis by the two methods were consistent.③ The oxidation and silver plating of dyed sections by the classical method are about 101 min,and the oxidation and silver plating of dyed slices by the modified method are about 34 min.Conclusion The improved PASM staining process staining time is less than 1 h,the staining background of the sections is clean,no impurities,the tissue structure is clear,the stability is good,the operation is convenient,and the feedback from pathologists is good,which is worthy of application and promotion in various pathology laboratories.
