The relationship between the expression of miR-195,LDH-5 and PTEN in tissues of patients with colorectal cancer and their clinicopathological parameters and prognosis

Abstract:Objective To investigate the relationship between the expression of micrornA-195(miR-195),lactate dehydrogenase-5(LDH-5),chromosome 10 deletion phosphatase and tensin homologous gene(PTEN)and pathological features as well as prognosis in patients with colorectal cancer.Methods 98 cases of colorectal cancer patients admitted to our hospital from March 2018 to March 2021 were selected,and the lesions and adjacent tissues were collected.The relative expression of miR-195 in tissues was determined by real-time reverse transcription polymerase chain reaction(qRT-PCR),and the expression of LDH-5 and PTEN in tissues was determined by immunohistochemistry.Receiver operating characteristic(ROC)curves were drawn to evaluate the area under the curve(AUC)of colorectal cancer,and to analyze the relationship between the expression of all three factors and the patient's general data,pathological features,and prognosis.Results The relative expression level of miR-195 and the positive rate of PTEN in colorectal cancer tissues were lower than those in paracancer tissues,and the positive rate of LDH-5 in cancer tissues was higher than that in paracancer tissues(P<0.05).The AUC(sensitivity and specificity)assessed by miR-195,LDH-5 and PTEN were 0.800(81.63%,75.51%),0.842(97.96%,70.41%)and 0.694(86.73%,52.04%).The AUC(sensitivity,specificity)of the three combined assessments was 0.954(90.82%,86.73%).The expression of miR-195 in cancer tissues of patients with stage Ⅲ to Ⅳ,low differentiation,lymph node metastasis,and distant metastasis was lower than that of patients with stage Ⅰ to Ⅱ,medium and high differentiation,no lymph node metastasis,and no distant metastasis(P<0.05).The positive rate of LDH-5 in patients with stage Ⅲ to Ⅳ,low differentiation,lymph node metastasis and distant metastasis was higher than that in patients with stage Ⅰ to Ⅱ,medium and high differentiation,no lymph node metastasis and no distant metastasis(P<0.05).The positive rate of PTEN in patients with lymph node metastasis and distant metastasis was lower than that in patients without lymph node metastasis and distant metastasis(P<0.05).The 3-year survival rate of the high miR-195 expression group was 76.19%,higher than that of the low expression group(49.35%);the 3-year survival rate of the LDH-5 negative group was 75.86%,higher than that of the positive group(46.38%);and the 3-year survival rate of the PTEN positive group was 65.96%,higher than that of the negative group(45.10%)(P<0.05).Conclusion In colorectal cancer tissues,the expression of miR-195 is down-regulated,the positive rate of LDH-5 is increased,and the positive rate of PTEN is decreased,all of which are related to lymph node metastasis and distant metastasis of colorectal cancer.Among them,miR-195 and LDH-5 are also related to TNM stage and differentiation degree.Patients with higher expression of miR-195,LDH-5 negative and PTEN positive had better prognosis.
