The expression of CCNA1,NFIB,and PREX2 in diffuse large B-cell lymphoma tissues and their relationship with clinical pathological features and prognosis

Abstract：Objective To analyze the expression of cyclin A1(CCNA1),nuclear factor I/B(NFIB),and phosphatidylinositol-3,4,5-triphosphate dependent Rac exchanger 2(PREX2)in diffuse large B-cell lymphoma(DLBCL),and their relationship with clinical pathological features and prognosis.Methods From February 2018 to February 2021,102 patients with DLBCL admitted to Handan Central Hospital were included as the observation subjects,and their clinical data,DLBCL tissues,and adjacent tissues were collected.According to the results of the 3-year follow-up study,patients were divided into two groups:64 cases in the survival group and 38 cases in the death group.Immunohistochemistry was used to detect the expression of CCNA1,NFIB,and PREX2 proteins in DLBCL tissues and adjacent non-cancerous tissues.Kaplan-Meier method was applied to analyze the relationship between CCNA1,NFIB,PREX2 expression and survival rate of DLBCL patients.Cox risk regression model was applied to analyze the prognostic factors of DLBCL patients.Results The positive expression rates of CCNA1,NFIB,and PREX2 in DLBCL tissues were higher than those in adjacent tissues(P<0.05).The expression of CCNA1,NFIB,and PREX2 proteins was associated with Ann Arbor staging,HANs typing,and Ki-67 positivity rate(P<0.05).The 3-year survival rates of CCNA1,NFIB,and PREX2 positive patients were lower than those of negative patients(χ2=7.726,χ2=16.980,χ2=6.981,P<0.05).Compared with the survival group,the death group had a higher proportion of patients with Ann Arbor stage III-IV,Non-GCB(HANs)typing,Ki-67 positivity greater than 70%,and positivity for CCNA1,NFIB,and PREX2 proteins.(P<0.05).Positive expression of CCNA1,NFIB,PREX2,and Ann Arbor stage III-IV were independent risk factors for mortality in DLBCL patients(P<0.05).Conclusion CCNA1,NFIB,and PREX2 proteins were found to be positively expressed at a higher rate in DLBCL tissues compared to paraneoplastic tissues.Their expression correlated with Ann Arbor staging,HANs typing,and Ki-67 positivity,suggesting their potential as prognostic clinical markers for DLBCL patients.
