Characterization and pre-staining mechanism of para-substituted benzene rings in methyl violet

Abstract:Objective To investigate the characteristics and advantages of the molecular structure of crystal violet in pre-staining of small tissues. Methods 60 cases of gastric endoscopic biopsy and puncture tissue samples were divided into 3 groups, each stained with eosin, hematoxylin, and crystal violet. After routine dehydration, the color intensity was observed with the naked eye. Infrared spectroscopy was used to detect the absorption peak characteristics of the three staining solutions, and the molecular structures were simulated using Materials Studio software. Results The staining effect of crystal violet was significantly better than that of eosin and hematoxylin; infrared spectroscopy showed that crystal violet has a special functional group, para-substituted benzene ring. The molecular structure simulation by Materials Studio software showed that crystal violet is a symmetric triphenylmethane molecule, and the triphenylmethyl radical in its structure is the key to producing the para-substituted benzene ring. Conclusion Compared with eosin and hematoxylin, crystal violet has a special functional group, para-substituted benzene ring, which makes it more stable in high temperature, high pressure, high concentration alcohol, and aldehyde environments, resulting in more vividly stained tissues after pre-staining.
