Abstract:Objective To explore the role of Congo red staining and oxidized Congo red staining in the diagnosis of amyloid light chain protein denaturation. Methods A total of 94 cases of amyloidosis diagnosed at Guangdong Provincial People's Hospital from January 2022 to January 2024 were collected, and they were subjected to Congo red staining, oxidized Congo red staining, and immunohistochemistry (EnVision method) for detection of κ and λ single light chain expression. Results The positive rate of Congo red staining was 100%, and the detection rate of primary protein (AL protein) by IHC was 89.4%. Among primary lesions, the AL protein positive rate was 100%, and all results were consistent with the diagnosis; oxidized Congo red staining detected AL protein in 87 cases, with 3 cases inconsistent with the diagnosis; there was no statistically significant difference between IHC and oxidized Congo red staining in terms of AL protein positive detection rate (χ2=0.58, P>0.05). Conclusion Histological Congo red staining and oxidized Congo red staining are simple and useful methods in the diagnosis of amyloid light chain protein denaturation, which can play a role in preliminary qualitative analysis, typing, and auxiliary verification of amyloidosis.
