Abstract:Objective To establish a method for periodic acid-Schiff (PAS) staining of semi-thin sections of epoxy resin-embedded muscle specimens, providing a more sensitive histological staining method for the pathological diagnosis of muscle glycogen storage disease (MGSD). Methods Retrospective analysis was performed on the pathological data of 18 patients with MGSD. Patients with atypical glycogen deposition characteristics were selected for PAS staining of semi-thin sections, and observations were made under light microscopy and electron microscopy. Results Conventional PAS staining showed that vacuoles within muscle fibers appeared as deep pink staining in semi-thin sections, indicating glycogen accumulation. Under electron microscopy, large amounts of glycogen particles were observed between myofibrils or within lysosomes, confirming abnormal glycogen deposition. Conclusion The PAS staining of epoxy resin semi-thin sections can effectively detect cases of glycogen accumulation that were missed by frozen section staining, providing a more sensitive histological staining method for the pathological diagnosis of muscle glycogen storage disease.
