Abstract:Objective To establish a practical method for preparing frozen sections of sural nerve biopsy tissue. Methods Using pathological plastic embedding boxes and self-made embedding auxiliary tools, we explored the freezing conditions and compared the quality of sectioning and staining to select the optimal freezing method for sural nerve biopsy tissue. Results Using the self-made embedding auxiliary tool, the nerve tissue sections were complete, with vertical cross-sections and no cracks after freezing for 30 seconds. Using the pathological plastic embedding box, the sections showed tissue fragmentation, defects, morphological changes, and inclined cross-sections. Conclusion This method is simple to operate, and the tissue structure is clear after staining. It can provide a nerve biopsy report within (0.52–2.5) hours, which is of great value for the rapid diagnosis of amyloid peripheral neuropathy and vasculitic peripheral neuropathy.
