Abstract:Objective To investigate the feasibility of using non-tissue specimens as positive controls in Ruhemann's copper staining. Methods A gel wax block was prepared using copper sulfate and gelatin, then sectioned and stained together with liver tissue specimens from patients with Wilson's disease. Results The copper sulfate-gelatin sections showed distinct staining, with a large number of brick-red copper particles observed under the microscope. Copper particle deposition was also visible in the liver tissue specimens from patients with Wilson's disease. Conclusion The sections prepared from copper sulfate-gelatin can meet the requirements for copper staining controls. They can be used as an indicator for staining when tissue specimens are difficult to obtain.
