Application of improved immunohistochemistry and elastic fiber dual staining in pathological diagnosis of pleural micrometastasis of non-small cell lung cancer

Abstract:Objective To evaluate the diagnostic value of pleural microinfiltration in non-small cell lung cancer,with using a dual staining method of immunohistochemistry AE1/AE3 and Gomori's aldehyde fuchsin staining for elastic fibers.Methods We collected 54 tissue samples from patients with non-small cell lung cancer and tumors adjacent to the pleura.Hematoxylin and eosin(HE)stain,staining of elastic fibers,and the dual staining of immunohistochemistry AE1/AE3 and elastic fibers were used to compare the diagnostic accuracy of these three staining method for determining pleural invasion accurately.Results Single staining with HE and single staining with elastic fibers yielded a diagnostic accuracy of 7%in determining pleural invasion.However,the dual staining of immunohistochemistry AE1/AE3 and elastic fibers achieved 100%accuracy in determining pleural invasion.Three staining method in ability for diagnosing pleural invasion accurately showed statistical differences(P<0.01).Conclusion The modified dual staining method,combining immunohistochemistry AE1/AE3 and Gomori's aldehyde fuchsin staining for elastic fibers,provides a superior assessment of tumor invasion into pleura compared to the single staining of HE and elastic fibers alone.This method enhances the precision of tumor staging,facilitating more accurate precision therapy.
