Abstract:Objective To investigate the expression of human aspartyl/asparaginyl β-hydroxylase(ASPH)in pleural fluid cell of patients with non-small cell lung cancer and its application value. Methods The pleural fluid samples of 80 NSCLC and 20 non-cancer patients were collected and made into cell wax blocks for detecting ASPH protein expression, compared with other commonly used lung cancer diagnostic biomarkers to analyze their relationship with clinical and pathological features,as well as their sensitivity and specificity for diagnosing NSCLC.Results The positive expression rate of ASPH in NSCLC pleural effusion cancer cells was higher than that in non cancerous pleural effusion(91.3%vs 0%,P<0.01),but there was no histological specificity. The positive rate of ASPH in the diagnosis of adenocarcinoma was higher than that of TTF1(90.9%vs 81.3%)and NapsinA(90.9%vs 77.5%),and the positive rate of ASPH in the diagnosis of squamous cell carcinoma was the same as that of P40(100%).The expression of ASPH in pleural effusion cells had statistical significance in different groups of tumor differentiation, lymph node metastasis and other organ metastasis(P<0.05),but there was no statistical difference in different groups of gender, age, smoking history, tumor size, pleural involvement and histological type. Conclusion ASPH is highly expressed in NSCLC pleural effusion cells, and combined with other classic diagnostic markers,it is helpful to improve the positive rate of pleural effusion cytology.High expression of ASPH is associated with poor prognosis, which has certain clinical application value.
