Application of polarized light microscope in histological staining of calcium oxalate crystals in rat kidney

Abstract:Objective To investigate the optimal detection effect of polarizer combined with histological staining on the renal tissues of rat a calcium oxalate stone model.Methods The FFPE tissues of 38 calcium oxalate stone model groups and 4 rat kidneys in the normal control group were 4 μm sections,and the presence state and birefringence of crystals and calcified crystals were used as evaluation indexes after staining with HE,Von Kossa,1%alizarin red S(PH4.2)and 0.2%alizarin red S(PH8.3).Results The crystals and calcium oxalate crystals of the model and control groups under the light microscope of the four staining were χ2=138.8,P<0.001,χ2=147.3,P<0.001;Under the polarizing microscopy of the four staining were χ ratio of 126.9,P<0.001,χ2 = 118.2,P<0.001,respectively.Von Kossa staining could determine the positive crystal and calcification crystal of the model group and the negative crystal of the control group,HE staining could only determine the crystal positivity of the model group and the negative of the control group,but could not determine the calcification crystal positivity of the model group and the negative of the control group.Both types of Alizarin red S did not stain on the crystal and show false negatives,making it impossible to determine;Since HE and alizarin red S staining failed to stain the crystals,the crystals with significant birefringence light under the polarizer microscope could determine the positive and no birefringence crystals in the model group and determine the negative of the control group,while the black granules or clumps in the Von Kossa staining were dark black or dark red under the polarizer,and the incomplete,scattered birefringence light but not could not significantly determine the positive crystals of the crystals and calcium oxalate.Conclusion Polarized light microscopy combined with HE staining can determine calcium oxalate crystals,Von Kossa staining can be used as a validation method for calcified crystals under light microscope,while alizarin red staining has no specificity for observing calcium oxalate crystals under light microscope.
