Expression of ALK fusion protein in triple negative breast carcinoma and its clinical significance

Abstract:Objective To investigate the ALK fusion protein expression and gene status in triple-negative breast carcinoma (TNBC).Methods The expression of ALK protein in 193 patients with TNBC and 20 cases with non-triple negative breast carcinoma (NTNBC)was detected by the Ventana immunohistochemistry (IHC) method.Then,the samples that had positive expression of protein were further examined with FISH and RT-PCR.Results There were 1.04% (2/193) ALK fusion protein positive cases among all of TNBC,which showed diffuse cytoplasmic granular coloration.The cytoplasmic and weak flaky coloration without granular sensation occurred in the focal tumor cells of 5.70% (11/193) TNBC.The nuclear/cytoplasmic coloration occurred in 11.40% (22/193) TNBC.Conclusions ALK gene rearrangement is not observed in TNBC.ALK fusion protein expression and copy number gain are only found in individual patient,though the correlation of them was unclear.
