Effects of Van-clear substituting xylene on HER2 protein expression in breast cancer

Abstract:Objective To compare the differences in the expression of HER2 protein in breast cancer between the environment-friendly transparent deparoffining solution Van-clear and xylene in immunohistochemistry,and to explore the feasibility of Van-clear instead of xylene.Methods 95 cases of breast invasive ductal carcinoma specimens were collected from the Zhongshan Boai Hosipital from June 2013 to March 2017.Two samples were taken from the same lesion site,and they were randomly divided into 2 groups,named Groups A and B.Xylene deparoffining was used in Group A,while Van-clear transparent deparoffining in Group B to make 95 slices.The general situation of the HER2 immunohistochemical staining was observed under the biological microscope,the expression of HER2 protein was detected by immunohistochemistry,and the difference of protein positive rate was compared between the two groups by SPSS20.0.Results Under the biological microscope,the results of the HER2 immunohistochemical staining in Groups A and B were as following:the background was clear,the positive staining was accurate (in the cell membrane),the positive intensity was reliable,the color was bright,there was no nonspecific staining,and the negative and positive control staining results were reliable.The positive rates of HER2 protein in Groups A and B were 24.21% and 26.32%,respectively.There was no significant difference in the positive rate between these two groups (x2 =0.50,P > 0.05).Besides,the coincidence rate of HER2 protein expression was 97.89%.Conclusion Environmental protection transparent deparoffining solution Van-clear has the potency to replace the traditional reagent xylene in the detection of HER2 protein expression in breast cancer by immunohistochemistry.
