Diagnostic roles of AP-2β, EGFR and P-cadherin in subtyping of pediatric rhabdomyosarcoma

Abstract:Objective To establish a panel of markers for pediatric rhabdomyosarcoma (RMS) histologic subgroup classification by immunohistochemistry.Methods 73 specimens of RMS were collected from Shanghai Children's Medical Center and Singapore KK Women and Children's Hospital.Immunohistochemical staining was performed by EnVision method using the markers AP-2β,P-cadherin,EGFR and myogenin;5-point scoring system was applied to calculate and analyze the results.Results Except 3 cases of spindle cell RMS,there were 70 patients selected,including 48 embryonal RMS (ERMS),22 alveolar RMS (ARMS),there was significant difference in the expression of AP-2β,P-cadherin and EGFR in ERMS and ARMS (P < 0.001),and no statistical significance in the expression of myogenin in ERMS and ARMS (P > 0.05).Conclusions The present panel of markers,including AP-2β,P-cadherin and EGFR may be useful in histologic subtype classification of RMS.
