Relationship among CCR4 expression, clinical pathological characteristics and prognosis in lung adenocarcinoma

Abstract:Objective To explore CCR4 expression in lung adenocarcinoma tissues and the relationship between the clinical pathological characteristics and prognosis.Methods The data of 629 patients of lung adenocarcinoma that did lung surgery and hospitalized in the Fourth Hospital of Hebei Medical University from March 2011 to March 2015 were collected;50 cases of normal lung tissue adjacent to carcinoma and 50 cases of pneumonia pseudotumor hyperplasia tissues were as control.Immunohistochemistry was used to detect the expression of CCR4 in lung adenocarcinoma and control groups.Real-time fluorescence PCR method in 629 cases of lung adenocarcinoma patients were used to detect the 18-21 EGFR exon gene mutation and then were followed up.Results The CCR4 expression in lung adenocarcinoma tissues were higher than in normal tissues adjacent to carcinoma and inflammatory pseudotumor hyperplasia tissues.Statistically significant difference was found in these groups(P < 0.05).The CCR4 expression was correlated with TNM stage,lymph node metastasis,pathologic grade,the size of the tumor,EGFR mutation and pleural invaded.Statistically significant difference was found in these groups (P < 0.05).The Kaplan-Meier survival analysis results showed that in CCR4 high expression groups of 12 months,24 months and 36 months cumulative survival was significantly lower than the low expression groups.COX multiple factor analysis results showed that EGFR mutation,high expression of CCR4 and the high grade were independent risk factors for the development of lung adenocarcinoma patients.Statistically significant difference was found in these groups (P < 0.05).Conclusions CCR4 is overexpressed in lung adenocarcinoma tissues,and its expression is related to TNM stage,lymph node metastasis,pathologic grade,the size of the tumor,EGFR mutation and pleural invaded.Patients with high expression of CCR4 have lower survival rate,and CCR4 is an independent risk factor for the survival of lung adenocarcinoma patients.
