Application of polyhydroxy acrylic acid in the detection of mycobacterium tuberculosis by Wade-Fite phenol alkaline magenta method

Abstract:Objective To study the practical value of the environmentally friendly fixative polyhydroxy acrylic acid in the detection of mycobacterium tuberculosis by Wade-Fite phenol alkaline magenta method.Methods 96 cases of tuberculosis lesions were collected from Zhongshan Boai Hosipital and Zhongshan People's Hospital from February 2013 to January 2017.2 specimens were obtained from the same lesion site and randomly divided into Groups A and B by lottery method.Group A used 4% neutral buffered formalin fixed to make 96 sliccs;while Group B used polyhydroxy acrylic acid fixed to make 96 slices.Both of them used Wade-Fite phenol alkaline magenta method to detect the mycobacterium tuberculosis.After that,SPSS20.0 was used to compare and analyze the different positive rates of mycobacterium tuberculosis in Group A and B.Results Under the biological microscope,Groups A and B were stained with the following results:both backgrounds were clean;the cell outlineswere clear,the nucleiwere blue,and the background coloring was light blue.Besides,the mycobacterium tuberculosis was bright red and its contrast with the background was good.The positive rates of the mycobacterium tuberculosis in groups A and B were 58.33% and 66.67% respectively,and these different positive rates of the two groups were statistically significant.(x2 =6.125,P < 0.05).Conclusion Environmental protection fixed solution polyhydroxy acrylic acid has the potential to replace the traditional reagent 4% neutral buffer formaldehyde applied to the detection of the mycobacterium tuberculosis by Wade-Fitephenol alkaline magenta method.
