Expression and significance of SASH1 and p-ERK in gastric adenocarcinoma

Abstract:Purpose To investigate the expression and significance of SASH1,a candidate tumor suppressor gene,and phosphorylated extracellular signal-regulated kinase (p-ERK1/2) in gastric adenocarcinoma,and to explore the relationship between these proteins.Methods The expression of SASH1 and p-ERK was immunohistochemically detected in 152 cases of gastric adenocarcinoma and 30 cases of adjacent tissues.The relationship between SASH1,p-ERK and the clinicopathological characteristics of gastric carcinoma and the correlation between the expression of SASH1 and p-ERK were analyzed.Results The positive rate of SASH1 expression was significantly lower in gastric carcinoma than that in adjacent tissues (P < 0.05),and significantly lower in poorly differentiated gastric adenocarcinoma than that in well-moderately differentiated gastric adenocarcinoma (P < 0.05).The positive rate of SASH1 expression was significantly lower in patients with Ⅲ-Ⅳ phase gastric cancer,having lymph node metastasis and having distant metastasis than those having Ⅰ-Ⅱ phase gastric cancer,without lymph node metastasis and without distant metastasis,respectively (P < 0.05;P < 0.01;P < 0.05).The positive rate of p-ERK1/2 expression was significantly higher in gastric carcinoma compared with that in adjacent tissues (P < 0.01),and significantly higher in poorly differentiated gastric adenocarcinoma than that in well-moderately differentiated gastric adenocarcinoma (P < 0.01).The positive rate of SASH1 expression was significantly higher in patients with Ⅲ-Ⅳ phase gastric cancer,having lymph node metastasis and having distant metastasis than those having Ⅰ-Ⅱ phase gastric cancer,without lymph node metastasis and without distant metastasis,respectively (P <0.05;P <0.05;P < 0.01).Significantly,the expression of SASH1 was inversely correlated with the expression of p-ERK in gastric adenocarcinoma (P < 0.05).Conclusion SASH1 may inhibit the progression of gastric carcinoma via mediating the signaling pathway of ERK1/2.
