Expression of immunohistochemical markers SALL4, CD30, D2-40 and glypican-3 and their utility in primary ovarian germ cell tumors

Abstract：Objective To investigate the expression and diagnostic value of the immunohitochemical markers SALL4,CD30,D2-40 and glypican-3 in primary ovarian germ cell tumors(OGCTs).Methods Immunohistochemical EnVision method was used to detect the expression of SALL4,CD30,D2-40 and Glypican-3 protein in 55 cases of primary ovarian germ cell tumors,including 16 dysgerminomas,10 embryonal carcinomas (ECs),12 yolk sac tumors (YSTs),1 choriocarcinoma,16 teratomas (6 immature,10 mature teratomas).10 normal ovarian tissues and 5 clear cell carcinomas were selected as control.Results All of dysgerminomas,ECs and YSTs were diffusely and strongly positive for SALL4.Focal SALL4 staining was seen in choriocarcinoma,half of the immature teratomas and 2 of 10 mature teratomas.CD30 was positive in ECs (8/10).However,the whole of dysgerninomas,YSTs,choriocarcinoma and teatomas were negtive staining.All of dysgernimomas showed diffusely and strongly D2-40 staining.ECs (3/10) were focal positive for D2-40,while YSTs,choriocarcinoma,teratomas were negative for D2-40.glypican-3 was diffusely positive in YSTs (11/12),focally weak in ECs (2/10) and choriocarcinoma (1/1),dysgerninomas and teratomas were negative for glypican-3.In contast,10 normal ovarian tissues and 5 clear cell carcinomas showed no SALL4,CD30,D2-40 and Glypican-3 staining.Conclusion SALL4 is a useful diagnosic marker for dysgerninomas;CD30,D2-40 and glypican-3 are good antibodies for diagnosis of ECs,dysgerninomas and YSTs,especially in differential diagnosis from clear cell carcinoma.
