MiR-423-5p promotes the apoptosis of lung cancer cells by targeting XIAP

Abstract:Objective To investigate the role and underlying mechanisms of miR-423-5p on apoptosis in lung cancer cells.Methods TCGA database was used to analyze the differences of miR-423-5p expression between tumor and normal tissues and its relationship with the prognosis of lung cancer.In the lung cancer cell lines A549 and PC-9,miR-NC,miR-423-5p and miR-423-5p inhibitor were transfected respectively to form miR-NC group,miR-423-5p group and miR-423-5p inhibitor group.Flow cytometry was used to detect the apoptosis of cells in each group.Dual-luciferase reporter assay was employed to verify the target gene of miR-423-5p.The relative expression levels of miR-423-5p and target gene mRNA in each group were detected by qRT-PCR.The protein expression levels of target gene in each group were detected by Western blotting.Results The expression level of miR-423-5p was significantly lower in lung cancer tissues than that in normal tissues(P<0.05),and high expression of miR-423-5p was positively correlated with patient prognosis(P<0.05).Compared with the control group(miR-NC group),overexpression of miR-423-5p(miR-423-5p group)led to a significant increase in apoptosis of lung cancer cells(P<0.05),whereas inhibition of miR-423-5p(miR-423-5p inhibitor group)led to a significant decrease in apoptosis of lung cancer cells(P<0.01).Dual-luciferase reporter assay demonstrated that,compared with the control group,miR-423-5p overexpression significantly inhibited the luciferase activity of the X-linked inhibitor of apoptosis protein(XIAP)3' untranslated region plasmid(P<0.01).The mRNA and protein expression levels of XIAP were significantly decreased in the miR-423-5p group compared with the control group(P<0.01),whereas they were significantly increased in the miR-423-5p inhibitor group(P<0.01).Conclusion miR-423-5p promotes lung cancer cell apoptosis by targeting XIAP,thereby inhibiting lung cancer progression.
