Abstract:Traumatic brain injury(TBI)is one of the leading causes of mortality and morbidity globally, often stemming from mechanical forces that induce primary injury, resulting in loss of consciousness, brain dysfunction,or even death.Mitochondria,as dynamic organelles responsible for cellular energy production, are integral to regulating energy, calcium homeostasis, signal transduction, and determining cell survival and fate during injury and pathological conditions.Mitochondrial dysfunction is a critical factor in secondary brain injury after TBI,characterized by an overproduction of reactive oxygen species and calcium overload, both of which significantly contribute to neuronal damage and are central to the mechanisms of secondary injury. In this review,we summarize the mitochondrial dysfunction associated with TBI and the various mechanisms involved.Furthermore,we discuss the advancements in a novel therapeutic strategy-mitochondrial transplantation for TBI-with the aim of offering a theoretical reference for clinical practice.
