Inhibitory effect and potential mechanism of Perceptide-TJ-2,an antimicrobial peptide designed based on artificial intelligence,on Hantaan virus infection

Abstract:Objective To investigate the inhibitory effect and primary antiviral mechanism of the artificial intelligence-designed antimicrobial peptides(AMPs)Perceptide-TJ-2 on Hantaan virus(HTNV)infection.Methods Immunofluorescence,qRT-PCR,Western blotting,and other techniques were employed to detect the virus expression following treatment with Perceptide-TJ-2 at various infection time points.The involvement of Perceptide-TJ-2 in early HTNV infection stages,such as attachment and penetration,was validated using the aforementioned methods.Additionally,surface plasmon resonance experiments were conducted to confirm the interaction between Perceptide-TJ-2 and HTNV Gn protein,while molecular docking simulations were utilized to elucidate the binding mechanism between Perceptide-TJ-2 and HTNV Gn.Results Pre-treatment of host cells or HTNV virus with the peptide before infection,or addition of the peptide and virus during infection,effectively inhibited HTNV infection.Subsequent analysis demonstrated that Perceptide-TJ-2 predominantly inhibited viral infection during the attachment stage by interacting with the HTNV Gn protein,thereby impeding viral entry into host cells.Conclusion The findings demonstrate the significant anti-HTNV infection activity of Perceptide-TJ-2,attributed to its ability to bind to HTNV Gn protein,thereby impeding virus-host cell interaction and exerting antiviral effects.This study provides a potential strategy for the development of novel AMPs-based drugs against HTNV infection.
