Abstract:Objective To establish an efficient multi-epitope vaccine design and development platform for emerging and sudden infectious disease pathogens to ensure the rapid design and preparation of vaccines with strong protective effects,good stability,and low side effects.Methods By predicting,screening(HLA binding capacity,immunogenicity,antigenicity,toxicity,sensitization,conservation,etc.)and optimizing the B-cell epitopes and T-cell epitopes of various subtypes of pathogens,the most immunoprotective and relatively conserved epitopes were selected to design universal multi-epitope combination vaccines targeting different mutant strains of pathogens.Results This platform was an efficient and universal platform for epitope vaccine design and development,integrating dominant antigen epitope prediction,property analysis,HLA binding capacity analysis,and multi-epitope vaccine structure prediction.It only required the sequence of pathogen antigenic proteins,and based on machine learning algorithms,a list of epitope sequences ranked from high to low score could be obtained,which could then be screened according to the properties of the epitope sequences,optimizing combinations to construct multi-epitope candidate vaccines with high population coverage,strong immunogenicity,and the ability to target multiple mutant strains.Conclusion The platform provides a universal vaccine research and development platform for a variety of pathogens,and provides a guarantee for the rapid design and preparation of vaccines with strong protective effects,good stability and low side effects.
