Efficacy of Bifidobacterium triple viable combined with agomelatine in treating female patients with depression and its effects on endocrine status,miR-195 and brain-derived neurotrophic factor

Abstract:Objective To observe the efficacy of Bifidobacterium triple viable combined with agomelatine in the treatment of female patients with depression and the effects on endocrine status,miR-195 and brain-derived neurotrophic factor(BDNF).Methods A total of 160 female patients with depression admitted from January 2023 to December 2024 were divided into two groups.The valdoxan group was given agomelatine treatment along with placebo treatment.The live bacteria group was treated with Bifidobacterium triple viable on top of agomelatine treatment.The two groups were compared in terms of 24-item Hamilton Depression Scale(HAMD)scores,miRNA relative expression,Treatment Emergent Symptom Scale(TESS),luteinizing hormone(LH),macrophage migration inhibitory factor(MIF),14-item Hamilton Anxiety Scale(HAMA)scores,follicle-stimulating hormone(FSH),BDNF,and nerve growth factor(NGF)levels.Results Compared with before treatment,there were significant differences in HAMD scores and HAMA scores between the two groups after treatment(P<0.05).After treatment,there were significant differences in the HAMD scores[(13.98±2.51)points vs(11.15±2.09)points]and HAMA scores[(9.36±2.05)points vs(7.41±1.69)points]between the valdoxan group and the live bacteria group,and the live bacteria group was lower than the valdoxan group(P<0.05).The total effective rate of the live bacteria group was 93.24%,which was higher than that of the valdoxan group(82.67%)(P<0.05).After treatment,the relative expression levels of miR-195 and miR-16 in the valdoxan group and the live bacteria group were higher than those before treatment,and the live bacteria group was higher than the valdoxan group(P<0.05).The relative expression levels of miR-124 in the valdoxan group and the live bacteria group after treatment were lower than those before treatment,and the live bacteria group was lower than the valdoxan group(P<0.05).There was no difference in LH and FSH before and after treatment between the two groups.Compared with before treatment,there were significant differences in BDNF,NGF and MIF between the two groups after treatment(P<0.05).After treatment,there were significant differences in BDNF[(17.87±3.15)μg/L vs(20.31±3.67)μg/L]and NGF[(18.68±3.12)ng/L vs(22.77±3.26)ng/L]between the valdoxan group and the live bacteria group,and the live bacteria group was higher than the valdoxan group(P<0.05).After treatment,the MIF in the valdoxan group and the live bacteria group[(57.11±8.63)μg/L vs(52.87±6.77)μg/L]was lower than that before treatment,and the MIF in the live bacteria group was lower than that in the valdoxan group(P<0.05).Conclusion Bifidobacterium triple viable combined with agomelatine for the treatment of depression in women improves depressive status,modulates BDNF levels as well as miRNA relative expression,and enhances efficacy.
