Study on the accelerated bone defect repair by pinoresinol diglucoside-fibrinogen complex

Abstract:Objective To explore the effect of local application of pinoresinol diglucoside(PDG)-fibrinogen complex on accelerating bone defect repair.Methods Forty-five SPF-grade Wistar rats were randomly divided into three groups:blank group(empty load,n=15),control group(fibrinogen-thrombin complex,n=15),and experimental group(PDG-fibrinogen complex,n=15).The experimental model of a mid-femoral segmental defect was performed in all rats.During the modeling process,the corresponding complexes were implanted into the bone defect areas respectively,and no further drug intervention was carried out after operation.X-ray films were taken every 2 weeks after operation to observe the repair of bone defects in each group at different time periods.At the 2nd,4th,and 8th weeks after operation,HE staining was performed on the bone tissues on the operated side to observe the repair of bone defects among different groups.At the 2nd week after operation,the expression of BMP2 in the early stage of bone growth in the bone defect areas of each group was observed by immunofluorescence staining.At the 4th week after operation,the growth of type H vessels in each group was observed by immunohistochemical staining.At the 8th week after operation,the new bone volume fraction and cortical bone thickness in the defect areas were observed according to the Micro-CT detection results.The three-point bending strength and flexural strength were observed by the three-point bending test.Results Imaging results and HE staining results showed that the repair speed of bone defects in the experimental group was significantly better than that in the blank group and the control group.The results of BMP2 immunofluorescence showed that the expression of BMP2 in the experimental group was higher than that in the other two groups.The immunohistochemical results showed that the expression level of type H vessels in the experimental group was higher than that in the other two groups.The biomechanical results showed that the three-point bending strength(P<0.05)and flexural strength(P<0.01)in the experimental group were better than those in the other two groups.The Micro-CT results showed that both the cortical bone thickness(P<0.05)and bone volume fraction(P<0.01)in the experimental group were better than those in the other two groups.Conclusion PDG-fibrinogen complex accelerates the repair of bone defects by upregulating the expression of BMP2 protein and promoting the formation of type H vessels.
