Abstract:Tumor growth depends on angiogenesis. With the increase of tumor volume, the degree of internal hypoxia exacerbates, which can stabilize hypoxia-inducible factors and upregulate the expression of various pro-angiogenic factors, thereby promoting endothelial cell activation and angiogenesis. The activation and expression level changes of signals and molecules such as VEGF,Notch, metabolic reprogramming and non-coding RNA are involved in this process.Based on the in-depth study of these molecular mechanisms, more and more intervention strategies such as antibody drugs, small molecule compound drugs, recombinant protein drugs, gene therapy and cell therapy have been developed, and the combination of these therapies with chemoradiotherapy and immunotherapy has shown good clinical application prospects, indicating that anti-angiogenic therapy is an important way for tumor treatment. This paper reviews the regulatory mechanisms of tumor angiogenesis and the research progress of anti-angiogenic therapy,in order to provide new ideas for the treatment of pathological angiogenesis diseases such as tumors.
