Establishment of a prognostic model for CD8+T cell infiltration,activation,and exhaustion in lung adenocarcinoma and role of the key gene ZNF540

Abstract:Objective To investigate the prognostic significance of CD8+T cell infiltration,activation,and exhaustion in lung adenocarcinoma(LUAD)and analyze the role of key gene.Methods A total of 41 CD8+T cell infiltration-activation-exhaustion genes(TIAEGs)were collected from TCGA and GEO databases.A prognostic model was constructed using LASSO regression,and the accuracy of the model was validated by Kaplan-Meier curves,random forest model,support vector machine model,ROC curves,and Cox regression analysis.The degree of immune cell infiltration was analyzed using the CIBERSORT algorithm.The expression difference of key protein ZNF540 was compared using the HPA database.KMPlotter database and Kaplan-Meier curves were used to analyze the impact of key genes on patient survival and immunotherapy.Results A prognostic model based on 10 TIAEGs was constructed.LUAD patients with higher TIAEGs scores had a poorer prognosis and were negatively correlated with immune cell infiltration.The key gene ZNF540 was significantly down-regulated in LUAD and positively correlated with the degree of immune cell infiltration and enrichment and immunotherapy response.Conclusion The TIAEGs score model and ZNF540 can serve as potential biomarkers for prognosis and immunotherapy response in LUAD.
