Epigenetic reprogramming mechanisms and intervention strategies of CD8+T cell exhaustion in tumor immune escape

Abstract:At present,tumor immunotherapy has become a powerful weapon in clinical tumor treatment.However,immune escape is a bottleneck that restricts the efficacy of tumor immunotherapy,and CD8+T cell exhaustion is one of the crucial causes in tumor immune escape.Researches have shown that CD8+T cells undergo epigenetic reprogramming under the influence of various immunosuppressive factors in the tumor microenvironment,which leads to CD8+T cell exhaustion.How to effectively target and regulate epigenetic reprogramming mechanisms to inhibit CD8+T cell exhaustion is currently a key and difficult problem that needs to be solved urgently.This paper reviews the characteristics of CD8+T cell exhaustion in the tumor microenvironment and epigenetic reprogramming mechanisms in tumor immune escape.It also elucidates the strategies for improving CD8+T cell exhaustion through targeted regulation of epigenetic mechanisms,and aims to provide new ideas and open up new pathways for tumor immunotherapy.
