Exploring the mechanism of adverse reactions caused by Tanreqing injection based on network pharmacology

Abstract:Objective To explore the molecular mechanism of adverse reactions caused by Tanreqing injection based on network pharmacology.Methods The compounds of Tanreqing injection were collected from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the potential targets were predicted in PharmMapper database and mapped with the related targets of adverse reactions in GeneCards database to obtain potential targets of Tanreqing injection causing adverse reactions.PPI network diagrams were constructed,and GO enrichment analysis and pathway analysis were performed on these targets to reveal the possible mechanism of adverse reactions caused by Tanreqing injection.Results A total of 44 compounds and 449 targets were screened by TCMSP,including 26 targets related to adverse reactions.The PPI network diagram showed that ALB,ACE,HMOX1,and PPARG were the four most-connected targets,involving biological processes such as response to exogenous stimuli,exogenous catabolic processes,and regulation of ERK1 and ERK2 cascade.The mechanism of action was related to the regulation of pathways in cancer,drug metabolism-cytochrome P450,metabolic pathways,and other biological pathways.Conclusion The main components of adverse reactions caused by Tanreqing injection include apigenin-7-O-glucoside,chrysin,apigenin,etc.ALB,ACE,HMOX1,and PPARG are its main targets of action.
