Expression analysis of genes related to cell junction and extracellular matrix in expanded skin regeneration

Abstract:Objective To analyze the expression of genes related to cell junction and extracellular matrix(ECM)in expanded skin by bioinformatic methods.Methods The high-throughput sequencing data of the expanded skin on rats scalp and the expanded skin on the back of mice were downloaded from GEO database,the differentially expressed genes(DEGs)were screened,the biological processes of DEGs sets in the two groups were analyzed by GO enrichment analysis and KEGG enrichment analysis,and the expression of genes related to cell junction and ECM were observed during expanded skin regeneration.In addition,CytoHubba was used to analyze Hub genes in expanded skin regeneration.Results There were 287 identical genes in the expanded scalp of the rat and the expanded skin of the mice back.GO analysis and KEGG analysis showed that the two DEGs sets and common DEGs were enriched in epidermal development,cell-cell junction tissue,cell-cell adhesion,ECM tissue,and IL-17 signaling pathway,among which 23 cell-cell junction genes were up-regulated in expanded skin,including adhesion junctions(Cdh2,Cdh3)and gap junctions(Gjb2,Gjb6),and 15 ECM genes were up-regulated in expanded skin,including Serpinb5,Klk7,Mpzl3,Tgm1,and Fermt1.Moreover,the common DEGs enriched in epidermal development,cell-cell junction tissue,ECM tissue,and cell-cell adhesion were characterized.Conclusion Through bioinformatic methods,we further understand the expression of genes related to cell junction and ECM in the process of expanded skin regeneration,providing a new way to study the mechanism of mechanical stretch-mediated expanded skin regeneration.
