Diagnostic value of chromosome 18 polysomy in malignant peripheral nerve sheath tumors and its correlation with clinicopathological features

Abstract:Objective To investigate the diagnostic value of chromosome 18 polysomy in malignant peripheral nerve sheath tumors(MPNST)and its correlation with clinicopathological features.Methods A total of 37 cases of MPNST diagnosed in Department of Pathology,Xijing Hospital from 2010 to 2024 were collected.Fluorescence in situ hybridization(FISH)was used to detect MPNST,and the clinicopathological features and immunophenotypes were summarized.Results Among the 37 patients,22 were males and 15 were females,with a mean age of 37 years(range:4~78 years).The mean tumor size was 10.8 cm.The most common primary site of MPNST was in the trunk(22/37).FISH results showed that 10 out of 37 cases(27.0%)presented with chromosome 18 polysomy.The statistical results showed that chromosome 18 polysomy was positively correlated with high mitotic activity(P=0.013)and tumor grade(P=0.005),and was more common in the head and neck(P=0.003).Conclusion In this study,FISH reveals chromosome 18 polysomy in MPNST,which may be correlated with a more aggressive histological morphology and higher grade of the tumor.It also provides a clue for the differential diagnosis with other spindle cell sarcomas.
