Ramipril Combined with ALT - 711 in Preventing Target Organ Remodeling in Spontaneously Hypertensive Rats

Abstract:Objective To compare the effects of Ramipril only and Ramipril combined with ALT - 711 on blood pressure and preventing target organ remodeling in spontaneously hypertensive rats(SHRs). Methods 24 male SHRs of 10 weeKs old were randomly divided into control group(group A),Rampril group(group B),ALT - 711 group (group C)and Rampril combined with ALT - 711 group(group D)with 6 SHRs in each group. SHRs in group A were gavaged with 1 ml/(Kg·d)normal saline. SHRs in group B were gavaged with 1 ml/(Kg·d)Rampril. SHRs in group C were gavaged with 10 mg/(Kg·d)ALT - 711. SHRs in group D were gavaged with 1 ml/(Kg·d)Rampril and 10 mg/(Kg·d)ALT - 711. Rat tail blood pressures were measured. Rats were Killed after 8 weeKs and right common carotid artery,heart and Kidney were acquired. Blood vessels,heart and Kidney remodelings were compared by tissue staining,immunohistochemical staining and image analysis. Collagen volume fraction,advanced glycation end products(AGEs)and fibronectin(FN)expression rates were measured. Results The blood pressure was the lowest in group D. SHRs in group D had the minimum arterial medial thicKness,the largest average profile area of the vessels and the least collagen content. Expression rates of AGEs and FN,left ventricular weight index,collagen con-tent,and Kidney weight index and expression of AGEs in Kidney were all the least among four groups. Conclusion Rampril combined with ALT - 711 can decrease blood pressure and prevent the remodeling of target organs more effec-tively.
