Effects of Oridonin to Inflammatory Factors of Endothelial Cells
Abstract:Objective To investigate the effects of oridonin to inflammation factors of human umbilical vein en-dothelial cells( HUVECs)induced by high glucose and high fat. Methods The model of HUVECs was established and divided into 4 groups:control group,high glucose and high fat group(33 μmol/L glucose and 500 μmol/L,48 h),2. 5μmol/L oridonin group and 5μmol/L oridonin group. The concentration of tumor necrosis factor-α( TNF-α)and interleukin-6(IL-6)in the supernatant were detected by enzyme-linked immunosorbent assay(ELISA)to observe the inflammation state. Results The concentrations of TNF-α as well as IL-6 in high glucose and high fat group were significantly higher than those in control group(t=14. 77,14. 05,P<0. 01). The concentrations of TNF-α(t=-4. 81,-9. 07,P<0. 01)as well as IL-6(t= -4. 23,-10. 06,P<0. 01)in 2. 5 μmol/L oridonin group and 5μmol/L oridonin group were significantly lower than those in high glucose and high fat group. Conclusion Oridonin can inhibit inflammation of HUVECs induced by high glucose and high fat.
