Fabrication and in vitro biological function of the nanofiber coating loaded SHED-Exo

Abstract:AIM:To fabricate a electrospun nanofiber coating composed of exosomes derived from stem cells of human deciduous exfoliated teeth(SHED-Exo)on the surface of titanium and to study the release profile,biocompatibility and biological function of the coating in vitro.METHODS:The ionic crosslinking method was used to prepare the Chitosan nanoparticles,incorporated with SHED-Exo(CS@Exo).And the nanoparticles were co-cultured and applied to the surface of titanium,which had been loaded the Polylactic Acid/Polyethylene Glycol(PLA/PEG)fibers through the electrospinning method to fabricate the PLA/PEG/CS@Exo coating.The release behavior of the coating was examined by PBS system in vitro.The biocompatibility was assessed using a cell co-culture method,and qRT-PCR was used to detect changes in the expression of related factors such as CD86,TNF-α,IL-10,and Arg-1.RESULTS:The nanoparticles showed good spherical geometry,and the average diameter of nanoparticles were(357.1±23.4)nm;the SHED-Exo incorporation efficiency was(59.3±2.8)%,respectively.The cumulative release rate was 84.3%in 30 days.The fibers coating showed rough and porous appearance.The drug-loaded microspheres were loaded onto the surface of the fibers.The cumulative release rate was 83.2%in 30 days.The cells were growing well on the fiber surface.The released SHED-Exo could be phagocytized by macrophages and promoted an increase in the expression of anti-inflammatory cytokines IL-10 and Arg-1(P<0.01).CONCLUSION:The fiber coating was a potential functional coating on implant surfaces to exert immunomodulatory effects.
