Abstract:AIM:Exploring the angiogenic effect of Xinjiang medicinal mulberry leaf extract on human umbilical vein endothelial cells(HUVECs)and providing theoretical basis for its potential application in periodontal tissue regeneration.METHODS:HUVECs were cultured in vitro to prepare Xinjiang medicinal mulberry leaf extract. The CCK-8 method was used to detect the effect of different concentrations of Xinjiang medicinal mulberry leaf extract on the proliferation activity of HUVECs.The appropriate concentration range for promoting HUVECs cell viability was screened and divided into blank control group and drug group(62.5,125 and 250 μg/mL mulberry leaf extract solutions).Flow cytometry was used to detect the apoptosis rate and reactive oxygen species expression levels of each group of cells, scratch assay and Transwell assay were used to detect cell migration ability, tubular formation assay was used to detect the in vitro tube forming ability of cells,RT-PCR was used to detect the mRNA expression levels of VEGF,bFGF,CD31 and CD34 in cells, and immunofluorescence staining was used to detect the expression of VEGF in cells.RESULTS:The cell viability was significantly enhanced at concentrations of 62.5,125 and 250 μg/mL mulberry leaf extract, with statistically significant differences(P<0.001).Compared with the blank group, mulberry leaf extract at different concentrations had no significant effect on cell apoptosis(P<0.05).The reactive oxygen species content of cells in each drug group decreased, and the relative horizontal migration rate, vertical migration rate, total length of in vitro tubular formation,VEGF,bFGF,CD31,CD34 mRNA and VEGF protein expression were significantly increased, with statistical significance(P<0.05).CONCLUSION:The extract of Xinjiang medicinal mulberry leaves can promote the proliferation, migration, and in vitro tubular formation of HUVECs,upregulate the expression of vascular formation related genes, reduce oxidative stress response, and thus promote the angiogenesis ability of HUVECs,which has potential effects on promoting periodontal tissue regeneration.
