A mendelian randomization study on the causal association between type 2 diabetes mellitus,hypertension,hyperlipidemia and oral lichen planus

Abstract:AIM:To investigate the causal relationship between type 2 diabetes mellitus,hypertension,hyperlipidemia and oral lichen planus(OLP)by Mendelian randomization(MR)analysis.METHODS:The data of the genome-wide association study(GWAS)for both the exposure group(type 2 diabetes mellitus,hypertension,and hyperlipidemia)and the outcome group(OLP)were sourced from the public database.From this dataset,the single nucleotide polymorphisms(SNPs)that exhibited a strong association with the exposure group were identified and utilized as instrumental variables,while those linked to confounders were excluded.The primary MR analysis method employed was the inverse variance weighted(IVW),and the stability of the results was subsequently verified through four sensitivity analyses.RESULTS:IVW results did not establish a causal relationship between type 2 diabetes mellitus,hypertension,hyperlipidemia and OLP[type 2 diabetes mellitus:OR=1.0000,95%CI(0.9213~1.0854,P=1.000);hypertension:OR=0.6944,95%CI(0.1681~2.8692,P=0.614);hyperlipidemia:OR=0.9736,95%CI(0.9030~1.0497,P=0.486)],and four sensitivity analysis did not reveal significant heterogeneity or horizontal pleiotropy in this result.CONCLUSION:No evidence was found to suggest a causal relationship between type 2 diabetes mellitus,hypertension,hyperlipidemia and OLP.
