FGF-2 promotes odotogenetic differentiation of dental pulp stem cells in 3D culture

Abstract:AIM:To investigate the effects of basic fibroblast growth factor(FGF-2) on odotogenetic dif-ferentiation of dental pulp stem cells (DPSCs) in 3D culture. METHODS: DPSCs were isolated and cultured from dental pulp tissue. The differentiation of DPSCs was assessed by osteogenic,odontogenetic and adipogenetic induction. Nanofibrous microspheres were synthesized, DPSCs were seeded onto the microspheres for 3D culture. When DPSCs were exposed to 20 ng/mL of FGF-2 for 7 d followed by 7 d-conventinal culture and 7 d-induction culture, the wdontogenetic differentiation related -gene and related -protein expression of DPSCs were detected by quantitative real-time PCR and immunocytochemistry respectively. RESULTS: Cultured DPSCs showed multiple differentiation potential. FGF-2 promoted odontogenetic differentiation related-gene (ALP, DMP1, OCN and DSPP) expression, especially after 2 w exposure(P<0.05). The increased expression of DSPP was found after 2 w exposure(P<0.05).CONCLUSION:FGF-2 may opromote dontoblast differentiation of DPSCs.
