Expression of Notch signaling related gens in human pulp cells stimulated by MTA

Abstract:AIM:To investigate the effects of mineral trioxide aggregate (MTA) on the expression of Notch1,Notch2 and Hes1 in human pulp cells (hDPCs).METHODS:hDPCs were in vitro cultured in the media with 0.2 mg/mL MTA,the mineralized nodule formation was observed by alizarin red staining.The mRNA expression levels of Notch1,Notch2,Hes1 and Runx2 were detected by quantitative RT-PCR.RESULTS:Cells grew normally in the media with 0.2 mg/mL MTA.MTA treatment produced a Von Kossa positive staining and the formation of mineralized nodules.The mRNA expression levels of Notch1 and Notch2 increased significantly on the 3rd day(P<0.05),then decreased gradually.The expression levels of Runx2 increased gradually over time,and on the 7th and 14th day were significantly higher than that of the control group (0 h) (P<0.05);whereas the expression of Hes1 was inbibited compared with the control group (0 h).CONCLUSION:Notch signaling may be involved in the mineralization regulation of hDPCs' stimulated by MTA.
