Licochalcone A induces the autophagy of human oral squamous cell carcinoma cells

Abstract:AIM:To investigate the effects of licochalcone A on the autophagy of human oral squamous cell carcinoma cells (OSCCs).METHODS:OSCC SCC-25 cells were treated once with 0,25 and 50 μmol/L of licochalcone A for 0,6 and 24 h respectively,and then the cells were harvested for further autophagy detection.The amounts of lysosomes were quantified by flow cytometry,and the gene and protein level of autophagy-related molecules were evaluated by real-time PCR,western blot and immunohistochemistry.RESULTS:Licochalcone A dose-and time dependently increased the autophagic activities of SCC-25 cells,characterized by the formation of lysosomes and autophagic vacuoles,increased gene and protein expression of beclin1 and LC3,and increased LC3-Ⅱ/LC3-Ⅰ ratio (P<0.05).No LC3 protain expression was observed in untreated SCC-25 cells,while after 50 μmol/L licochalcone A treatment high level of LC3 protein in SCC-25 cells was observed,predominantly in cytoplasm (P<0.05).CONCLUSION:Licochalcone A may promote the sautophagic activitiy of SCC-25 cells,which might be one of the important mechanisms of its anti-tumor effects.
