Local application of simvastatin suppresses inflammatory bone destruction via regulation of RANKL/OPG in periapical tissues

Abstract:AIM:To observe the effect of simvastatin on RANKL/OPG expression and bone destruction in perioapical tissues.METHODS:Apical periodontitis was induced in 8 -week -old male Wistar rats(n =50)by direct exposure of the dental pulp chamber of mandibular first molars to the oral cavity.Simvastatin solution (1 g/L )was in-jected under buccal periosteum of the tooth of 25 rats on 0,3,6,9,12,15,18 and 21 d postoperatively.The Animals of the 2 groups were randomly sacrificed on day 1,3,7,14 and 21 respectively after operation(n =5).RANKL/OPG expression jaw tissues were examined by immunohistochemical staining.The number of osteoclasts was measured by TRAP staining.RESULTS:Compared with control group,RANKL expression was significantly lower in simvastatin group on 3 and 21 postoperative day (P <0.05).OPG expression was significantly higher in simvastatin group at all time points except 1 day after eperation(P <0.05).RANKL/OPG ratio and the number of osteoclasts was significantly lower in simvastatin group(P <0.05).CONCLISION:Local applications of simvastatin can inhibit the formation of osteo-clasts through regulating the expression of RANKL and OPG.
