The osteogenic activity of adipose-derived stem cells isolated from type 2 diabetic rats in vitro

Abstract:AIM:To study the differentiation potency of adipose-derived stem cells (ASCs)from type 2 dia-betic (T2MD)rats in vitro.METHODS:16 male SPF SD rats aged 8 weeks were divided into 2 groups,8 rats were use for the etablishement of T2MD models,the other 8were fed with the routine diet as normal controls.Blood glucose levels were monitored.ASCs were isolated from the bilateral inguinal fat pads.Cell morphology was observed under phase con-trast microscope.Cell proliferation was tested by CCK-8 kit.The surface markers of the cells were examined by flow cy-tometry.Cell differentiation was evaluated by osteoinduction and adipoinduction.The mRNA levels of ALP,OCN,OPG and RUNX2 were analyzed by RT-PCR after 7-day osteoinductive culture.RESULTS:The morphology and proliferation abilities of ASCs of the 2 groups were not significantly different.Both groups of ASCs expressed highet CD44 and CD90, and lower CD34 and CD45.ASCs from diabetic rats had relatively low osteogenic potential (P<0.05)and relatively high adipogenic potential (P<0.05 )than the controls.The ALP activity was lower (P<0.05 ),the expressions of ALP, OCN,OPG and RUNX2 were down-regulated in diabetic group(P<0.05).CONCLUSION:The osteogenic differenti-ation potential of the ASCs from diabetic rats is lower thant that of the healthy SD rats in vitro.
