Effects of basic fibroblast growth factor on the biological charactor of gingiva-derived mesenchymal stem cells

Abstract:AIM:To investigate the effects of basic fibroblast growth factor ( bFGF) on the biological char-actor of gingiva-derived mesenchymal stem cells ( GMSCs) in vitro. METHODS:GMSCs were isolated from human healthy gingival tissue samples. Proliferation, colony formation, alkaline phosphatase ( ALP) activity, STRO-1 expres-sion, osteogenic and adipogenic differentiation of GMSCs cultured in the presence or absence of bFGF, were evaluated. RESULTS:Treatment with 10 ng/mL bFGF significantly increased the proliferation and STRO-1 expression of GM-SCs. bFGF also increased oil red O staining following adipogenic induction and the expression of PPARγ and LPL of GMSCs. In contrast, it reduced the ALP activity and mineral nodule formation;inhibited the gene expression of ALP, OCN and BSP of GMSCs. CONCLUSION:Under certain conditions, bFGF enhances GMSCs stemness by up-regula-ting stem cell gene expression, increasing proliferation ability, and may regulate the differentiation potency of GMSCs.
