Effect of tumor necrosis factor-αon bone marrow mesenchymal stem cell apoptosis

Abstract：AIM:To investigate the effects of tumor necrosis factor-α ( TNF-α ) on the apoptosis of rat bone marrow mesenchymal stem cells ( BMSCs) . METHODS:Mouse BMSCs were cultured and stimulated with TNF-α(μg/L)at 0, 20 and 40 respectively for 24 h. Cell activity staining and Tunel staining were used to determine the apoptosis of BMSCs. The expression of bax and Bcl-2 protein was examined by Western blot. RESULUTS: TNF-αdose dependently decreased the survival and increased the apoptosis of BMSCs. TNF-α up-regulated Bax expression and down-regulated bcl-2 expression (P<0. 05). CONCLUSION:TNF-α can induce the apoptosis of BMSCs.
