Construction of a HtrA deficient Streptococcus mutans strain and its transformation ability

Abstract：Abstract] AIM:To construct a HtrA deficient strain of Streptococcus mutans(S.mutans),and to compare the transformation ability of the deficient strain with HtrA high virulence strain.METHODS:S.mutans strain with HtrA high virulence was cultured in anaerobic condition and the competent cells were prepared.Recombinant plasmid vector as inserted into the cells and homologous recombination technique was used to construct the HtrA deficient strain of S.mutans.The constructed strain was identified by PCR method.The transformation ability of the 2 strains was in-vestigated by counting the number of colonies cultured on TPY medium for 6 -48 hours.RESULTS:The HtrA defi-cient strain was constructed.At all time points tested,the colony forming unit of the deficient strain was significantly lower than that of the high virulent strain (P <0.05).CONCLUSION:Knock-out of HtrA gene may reduce the transformation ability of S.mutans,indicating that HtrA is closely related to the virulence of S.mutans.
